| Description | In this work we present an optimized method for peptide separation and quantitation by capillary electrophoresis with mass spectrometry detection (CE-MS), which can be used for enzyme kinetics and inhibition assays of ß-secretase (BACE). It is a key enzyme involved in creation of amyloid senile plaques, which has been proposed to play essential role in pathogenesis of the Alzheimer’s disease. As a result the ongoing medical and pharmacological research is thus focused on the prevention of the production of beta-amyloid by inhibiting of BACE. The Fluorescence Resonance Energy Transfer based assays are used almost exclusively for this purpose, unfortunately these suffer from number of conceptual disadvantages. Employing CE-MS enables high sensitivity, low sample consumption and rapid analysis time, what is more no expensive labelled substrates are necessary. |
