Adiponectin gene variability and plasma levels in non-obese Czech patients with asthma

Popis Background and Aim: Adiponectin (ADIPOQ) gene variants and ADIPOQ circulating levels were previously associated with obesity and asthma risk. This pleiotropic cytokine, which modifies inflammation, is produced by the adipose tissue and T cells, as recently described. Obesity has a significant impact on asthma risk, phenotype, and prognosis. The aims of this study were to analyse ADIPOQ single nucleotide polymorphism (SNP) and ADIPOQ plasma levels in non-obese Czech patients with asthma and systematically healthy subjects, and thus to investigate the possible impact of ADIPOQ in the pathogenesis of asthma independently of obesity. Methods: In total, 576 non-obese individuals (with a body mass index less than 30) were included in this case-control study: 303 patients with asthma and 273 systematically healthy controls. Clinical examination was performed, IgE levels were determined and skin-prick tests to assess the reactivity for various allergens were evaluated. All subjects were genotyped for ADIPOQ (rs1501299) SNP by qPCR method with a TaqMan fluorescent assay. ADIPOQ plasma levels were determined by enzyme linked immunosorbent assay (ELISA) kit in subgroups of 46 patients with asthma and 25 healthy controls. Results: Significant differences in allele and genotype frequencies of ADIPOQ (rs1501299) SNP between patients with asthma and controls were detected (P<0.001); carriers of GG genotype vs. those with GT+TT genotypes had a higher risk of developing asthma (P<0.001, OR=1.88, 95%CI: 1.35- 2.63). Plasma levels of ADIPOQ were similar between patients with asthma and controls; and they were independent of ADIPOQ (rs1501299) individual´s genotype. Conclusions: This is the first study focusing on ADIPOQ gene variability and asthma in Caucasian population. We suggest that ADIPOQ (rs1501299) SNP may be a marker of asthma risk for non-obese individuals.
